Engineered Streptavidin Monomer and Dimer with Improved Stability and Function


Tetrameric Streptavidin's high affinity for biotin has been extensively utlized, however, the tetrameric structure may interfere with some assays. A stable monomer of Streptavidin was prepared with rationally designed mutations. The mutations improved the stability of the monomer. As expected, the affinity of the constructed dimer showed higher affinity towards biotin than the constructed monomer but was still much lower than WT Streptavidin. Nevertheless, this approach still present potential in future applications and elucidating the oligomerization in Streptavidin function.
